Genetic investigation makes it possible to identify them<\/h2>\n<p style=\"text-align: center;\">by Matteo Cavallito<\/p>\n<p>&nbsp;<\/p>\n<p>Soil <strong>microorganisms<\/strong> could be an underestimated source of new antibiotic production. This is reported in a <a href=\"https:\/\/scholarlypublications.universiteitleiden.nl\/handle\/1887\/4082475\" target=\"_blank\" rel=\"noopener\">study<\/a> by the <strong>Leiden Institute of Biology<\/strong> in the Netherlands. The research thus highlights new development opportunities for these compounds, which are of particular importance in the face of known problems with pathogen <a href=\"https:\/\/resoilfoundation.org\/en\/articles\/microbes-soil-antibiotic-resistance\/\" target=\"_blank\" rel=\"noopener\">resistance<\/a>. The search for new antibiotics \u201chas slowed significantly, despite the urgent need for solutions,\u201d a <a href=\"https:\/\/www.universiteitleiden.nl\/en\/news\/2024\/11\/the-solution-to-antibiotic-resistance-might-be-under-our-feet\" target=\"_blank\" rel=\"noopener\">statement<\/a> says. To date, <strong>about 70 percent of these products<\/strong> in use date back to the 1950s, the industry&#8217;s decade of greatest expansion that was followed by a long decline in discoveries of new bactericides that continues to this day.<\/p>\n<p>&nbsp;<\/p>\n<div class='avia-iframe-wrap'>\n<blockquote class=\"wp-embedded-content\" data-secret=\"641dKPsrLl\"><p><a href=\"https:\/\/resoilfoundation.org\/en\/articles\/microbes-soil-desorption\/\">Microbes are resurging in decontaminated soils thanks to desorption<\/a><\/p><\/blockquote>\n<p><iframe loading=\"lazy\" class=\"wp-embedded-content\" sandbox=\"allow-scripts\" security=\"restricted\" style=\"position: absolute; visibility: hidden;\" title=\"&#8220;Microbes are resurging in decontaminated soils thanks to desorption&#8221; &#8212; Re Soil Foundation\" src=\"https:\/\/resoilfoundation.org\/en\/articles\/microbes-soil-desorption\/embed\/#?secret=cjaOXPxBcg#?secret=641dKPsrLl\" data-secret=\"641dKPsrLl\" width=\"600\" height=\"338\" frameborder=\"0\" marginwidth=\"0\" marginheight=\"0\" scrolling=\"no\"><\/iframe><\/div>\n<h5>Soil bacteria are an untapped resource<\/h5>\n<p>\u201c<strong>Antibiotic-resistant bacteria<\/strong> pose a major health threat,\u201d the research states. \u201cAddressing this challenge requires among others the development of new antibacterial compounds.\u201d Today many new and potentially more potent antibiotics are still waiting to be discovered, points out <strong>Nataliia Machushynets<\/strong>, a biologist and author of the study highlighting the role of microorganisms in producing these compounds that can kill bacteria.<\/p>\n<blockquote><p>The research focuses \u201con discovering novel antibiotics from soil bacteria, specifically <strong>Streptomyces<\/strong> and <strong>Paenibacillus<\/strong>. Despite the vast potential encoded within <strong>bacterial genomes<\/strong>, only a fraction of the natural products have been explored experimentally.\u201d<\/p><\/blockquote>\n<p>Identifying promising new compounds remains a complex task that requires the use of new and improved techniques. Genomic analysis through <strong>DNA sequencing<\/strong> is one of them. Using these tools \u201cmade us realise we\u2019ve barely scratched the surface of what bacteria are capable of producing,\u201d Machushynets explains. \u201cCurrent estimates suggest we know<strong> less than 5 percent<\/strong> of their potential.\u201d<\/p>\n<div class='avia-iframe-wrap'>\n<blockquote class=\"wp-embedded-content\" data-secret=\"pRGNVBdObc\"><p><a href=\"https:\/\/resoilfoundation.org\/en\/articles\/china-antibiotic-resistance-soil\/\">Chinese scientists provide first global map of antibiotic resistance in soil<\/a><\/p><\/blockquote>\n<p><iframe loading=\"lazy\" class=\"wp-embedded-content\" sandbox=\"allow-scripts\" security=\"restricted\" style=\"position: absolute; visibility: hidden;\" title=\"&#8220;Chinese scientists provide first global map of antibiotic resistance in soil&#8221; &#8212; Re Soil Foundation\" src=\"https:\/\/resoilfoundation.org\/en\/articles\/china-antibiotic-resistance-soil\/embed\/#?secret=T5bpyXkK4r#?secret=pRGNVBdObc\" data-secret=\"pRGNVBdObc\" width=\"600\" height=\"338\" frameborder=\"0\" marginwidth=\"0\" marginheight=\"0\" scrolling=\"no\"><\/iframe><\/div>\n<h5>Identifying compounds with metabolomics<\/h5>\n<p>Currently, the study further points out, \u201ca critical obstacle in antibiotic discovery is the <strong>repeated rediscovery of known compounds<\/strong>, a process called replication.\u201d To solve this problem, the researchers adopted a technique called <a href=\"https:\/\/en.wikipedia.org\/wiki\/Metabolomics\" target=\"_blank\" rel=\"noopener\">metabolomics<\/a>, which involves studying the so-called chemical fingerprints of cellular processes. In this way they were able to identify promising compounds in the samples they examined.<\/p>\n<p>Specifically, the techniques used were made available in a platform called nanoRAPIDS, designed, the study further explains, \u201cfor the discovery and <strong>dereplication<\/strong> of low-abundance bioactive compounds through at-line nanofractionation, molecular networking, and bioactivity assays.\u201d Such a platform \u201cwas successfully applied to analyze the bioactive crude extracts of Bacillus and Streptomyces and highlighting the individual mass features that contribute to the activity in the crude extract.\u201d<\/p>\n<div class='avia-iframe-wrap'>\n<blockquote class=\"wp-embedded-content\" data-secret=\"SQ3REE9UlK\"><p><a href=\"https:\/\/resoilfoundation.org\/en\/articles\/protecting-forests-respiratory-diseases\/\">Protecting reduces respiratory diseases, a study has found<\/a><\/p><\/blockquote>\n<p><iframe loading=\"lazy\" class=\"wp-embedded-content\" sandbox=\"allow-scripts\" security=\"restricted\" style=\"position: absolute; visibility: hidden;\" title=\"&#8220;Protecting reduces respiratory diseases, a study has found&#8221; &#8212; Re Soil Foundation\" src=\"https:\/\/resoilfoundation.org\/en\/articles\/protecting-forests-respiratory-diseases\/embed\/#?secret=xc5CTo2ctc#?secret=SQ3REE9UlK\" data-secret=\"SQ3REE9UlK\" width=\"600\" height=\"338\" frameborder=\"0\" marginwidth=\"0\" marginheight=\"0\" scrolling=\"no\"><\/iframe><\/div>\n<h5>New frontiers for antibiotics<\/h5>\n<p>While advanced spectroscopic techniques, such as <strong>metabolomics<\/strong>, have made it possible to detect and characterize antibiotics present in complex bacterial extracts, remember the study, synthetic chemical analysis and bioinformatics have helped researchers discover, optimize and refine the <strong>antibacterial properties<\/strong> of compounds. Among the antibiotics identified were tridecaptin, effective against a bacterium known as Pseudomonas aeruginosa and resistant to colistin, and bioactive paenitracin, which acts against vancomycin-resistant Enterococcus faecium.<\/p>\n<blockquote><p>Other <strong>compounds identified<\/strong> included paenylipoeptins, quinazolinones, actinomycin L and tridecaptin lipopeptides, and paenitracin, a new bacitracin variant.<\/p><\/blockquote>\n<p>The research, in short, used \u201ca wide range of computational, biological, and analytical chemical approaches to activate cryptic biosynthetic pathways and find new chemistry, thus contributing to the need for new chemical space as the basis for our future antibiotics.\u201d The findings, the study concludes, \u201chighlight that there still is a lot of potential in soil bacteria as sources of novel antibiotics, to aid us in the continuing combat against multi-resistant bacterial infections.\u201d<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Soil, a Dutch study recalls, harbors a huge number of microbes capable of producing effective and as yet unknown antibiotics.
